Human IL-4 ELISA

Interleukin-4 (IL-4) is a cytokine that that plays many biological roles, including the stimulation of activated B-cell and T-cell proliferation, and the differentiation of CD4+ T-cells into Th2 cells. It is a key regulator in humoral and adaptive immunity. IL-4 binds to a cell surface receptor complex that consists of the IL-4 binding protein (IL-4R alpha) and the gamma chain of the IL-2 receptor complex (gamma c).

B. Principle of the technology

IL-4 ELISA is based on the principle of a solid phase enzyme-linked immunosorbent assay. The assay utilizes rabbit anti-human IL-4 for immobilization on the microtiter wells and biotinated rabbit anti-human IL-4 antibodies along with streptavidin conjugated to horseradish peroxidase (HRP) for detection. The test sample is allowed to react simultaneously with the two antibodies, resulting in the IL-4 molecules being sandwiched between the solid phase and enzyme-linked antibodies. After incubation, the wells are washed to remove unbound-labeled antibodies. A HRP substrate, TMB, is added to result in the development of a blue color. The color development is then stopped with the addition of Stop Solution changing the color to yellow. The concentration of IL-4 is directly proportional to the color intensity of the test sample. Absorbance is measured spectrophotometrically at 450 nm.
