STAT3 Filter Plate AssaySTAT3 protein plays an important role in development, immune system and inflammation. When activated, STAT3 translocates from cytoplasm to nucleus, binds to the STAT3 responsive element and regulates the expression of genes, involving in cell growth, division, transformation and apoptosis. Dysfunction of STAT3 is closely associated with many diseases, such as cancers. Constitutively activated forms of STAT3 have been detected in many cancer cell lines and primary tumors. B. Principle of the technology Signosis’ STAT3 filter assay is a plate-based analysis for monitoring the activity of STAT3. In the assay, biotin labeled STAT3 DNA binding sequence is mixed with nuclear extract to allow formation of STAT3-DNA complex. A filter plate is used to retain bound STAT3 probe and remove free DNA probe. The bound pre-labeled STAT3 probe is then eluted from the filter and hybridize to the corresponding well of 96 well of Hybridization Plate for quantitative analysis. The captured STAT3 probe is further detected with streptavidin-HRP. Luminescence is reported as relative light units (RLUs) on a microplate luminometer. Figure 1
