YY1 Filter Plate Assay

YY1 plays a fundamental role in normal biologic processes, such as embryogenesis, differentiation, replication, and cellular proliferation. YY1 exerts its effects on genes involved in these processes via binding to YY1 recognition site (the consensus sequence) of the promoter regions. The dysfunction of YY1 is associated with abnormal cellular proliferation, apoptosis, tumorigenesis and metastasis. Signosis has developed YY1 filter plate assay for monitoring DNA binding activity of YY1, which can analyze multiple samples simultaneously.

B. Principle of the technology

Signosis’ YY1 filter assay is a plate-based analysis for monitoring the activity of YY1. In the assay, biotin labeled YY1 DNA binding sequence is mixed with nuclear extract to allow formation of YY1-DNA complex. A filter plate is used to retain bound YY1 probe and remove free DNA probe. The bound pre-labeled YY1 probe is then eluted from the filter and hybridize to the corresponding well of 96 well of Hybridization Plate for quantitative analysis. The captured YY1 probe is further detected with streptavidin-HRP. Luminescence is reported as relative light units (RLUs) on a microplate luminometer.

Figure2. YY1 filter plate assay analysis. HeLa cells were starved with serum-free medium for 8 hours, and treated without and with 20ng/ml PMA for 30 minutes. The nuclear extracts were prepared, and subjected to YY1 filter plate assay.
