Transcription factors (TFs) are a group of cellular proteins that play essential roles in regulating gene expression. They act as sensors to monitor cellular change and convert the signals into altered gene expression. Often, a specific cellular signal pathway can activate multiple TFs and the expression of a specific gene is under control of multiple TFs. Hence, monitoring the activation of multiple TFs simultaneously is critical to understand the molecular mechanism of cellular regulation underlying cell signaling and gene expression. Lysosomal Stress TF Activation Profiling Plate Array FA-1012


The lysosome plays a key role in cellular homeostasis by controlling both cellular clearance and energy production to respond to environmental cues. It is well known that transcription factor EB (TFEB) and mTOR complex 1 (mTORC1) are master regulators of lysosomal biogenesis. Under starvation, cells initiate the process of autophagy and increase the capacity of the lysosome by the lysosome stress response. mTORC1 dissociates from the lysosome and becomes inactivated, leading to dephosphorylation of TFEB and autophagy. Dephosphorylated TFEB enters the nucleus, binds to the enhancer element CLEAR, and activates transcription of genes related to the lysosome as well as autophagy. The lysosome stress response is closely related to various diseases such as Huntington’s disease, liver disease and lysosomal storage diseases including Pompe disease. Recently, more TFs have been reported to be associated with lysosome stress, such as p53, FOXO1, PPAR, Stat3, NFkB, HIF, HSF and MITF.

Stem Cell TF Activation Profiling Plate Array I FA-1003


Stem cells are important cells characterized by their ability to self-renew or differentiate into many cell types, which is governed by intracellular signaling pathway and transcription regulation. Hence, transcription regulation plays a determinant role in conferring cellular identity and function. The step-wise maturation of stem cells into terminally differentiated cell types requires the timely activation of a cascade of transcription programs governed by lineage-specifying transcription factors. Transcriptional regulation plays a critical role in the control of biological processes, and transcription factors have been considered as master switches for cell fate determination. The cancer stem cell paradigm postulates that decontrolled tissue-specific stem cells or progenitor cells are precursors for cancer biogenesis. Cancer stem cells are a subset of cancer cells within a tumor that have stem cell-like characteristics. During tumorigenesis, deregulated transcription factor expression or activation can promote abnormal self-renewal, proliferation, and differentiation of the cells. Cholesterol is an essential component of cell membranes to ensure cell viability and cell growth, and also serves as a precursor for steroids and bile acids. The changes in cholesterol level could lead to abnormal biological process.The elevated levels have been reported to be associated with several diseases, including cardiovascular disease, heart attack, stroke and Huntington's disease. Many metals and metal-containing compounds including chromium (Cr), nickel (Ni) have been identified to be potent mutagens and carcinogens in human and animals. Metal can disrupt homeostasis by generating oxidative stress, impairing DNA repair and disrupting protein binding and normal cellular function. Metals can directly induce gene expression by activating metal-responsive factors and signal transduction pathways, such as MAPK/RAS/Raf, PI3K/AKT, JAK/STAT and IKK/Ikb/NFkB pathways.  It has been reported that the cell cycle arrest is a passive cellular defense mechanism in response to external metal toxicants, and improper cell growth regulation may results in cancer development.
