TF Filter Plate Assay without Probe FA-0010


Transcription factors (TFs) are a group of cellular proteins that play essential roles in regulating gene expression. They act as a sensor to monitor cellular change and convert the signals into altered gene expression. A specific TF is often regulated by multiple upstream proteins. Monitoring the activation of a TF is critical to uncovering the mechanism of signal pathways and cellular regulations.

To perform the assay, a TF Filter Assay Kit and a probe are needed.

NFkB Filter Plate Assay FA-0001


Nuclear factor-kB (NFκB) is a transcription factor that controls the expression of genes involved in immune responses, apoptosis, and cell cycle. NFκB can be activated by a variety of stimuli, including cytokines, T and B cell mitogens, viral proteins, and stress inducers. The stimulation leads to phosphorylation of its inhibitor IκB, which releases NF-κB from IkB complex. After dissociation, NFκB translocates from the cytoplasm into the nucleus, where it binds to its target DNA elements and positively regulates the transcription of its target genes. Monitoring the activity of NFκB is crucial to the studies of the mechanism underlying NFκB activation and its associated pathway. HIF Filter Plate Assay FA-0002


Hypoxia-inducible factor (HIF) has been recognized primarily for its role in the maintenance of oxygen and energy homoeostasis. HIF-1 and HIF-2 are heterodimeric transcription factors composed of HIF-1 and HIF-2 , respectively, which dimerize with a constitutively expressed ß subunit and subsequently bind to hypoxia response elements in the promoters of target genes. The activity of HIF is regulated by a variety of stimuli, including changes in cellular oxygen concentration, growth factors and oncogenic activation. Signosis has developed HIF filter assay. Unlike conventional gel shift assays, HIF filter assay is a high throughput assay and it can analyze DNA binding activities of HIF in multiple samples simultaneously. ER Filter Plate Assay FA-0003


ERs (ERalpha and ERbeta) mediate cellular signaling of estrogens, belonging to the nuclear receptor family of transcription factors. The mechanism of regulatory actions of ERs is that ligand-bound ERs bind directly to estrogen response elements (ERE), in the promoters of target genes or via protein-protein interactions with other transcription factors. The dysfunction of ERs has been demonstrated to be closely related with breast cancer, prostate cancer and lung cancer. AP1 Filter Plate Assay FA-0004


AP-1, a dimeric protein complex composed of Jun and Fos proteins, plays a key role in the control of cell death, proliferation and transformation. Ap1 protein can be activated by many stimuli, including TPA, serum, growth factors and oncoproteins, TNF and IL-1, as well as by a variety of environmental stresses, such as UV radiation. When it is activated, AP1 protein binds to AP- DNA recognition element within the promoter regions of target genes and regulates the gene expression. STAT3 protein plays an important role in development, immune system and inflammation. When activated, STAT3 translocates from cytoplasm to nucleus, binds to the STAT3 responsive element and regulates the expression of genes, involving in cell growth, division, transformation and apoptosis. NRF2 Filter Plate Assay FA-0007


Nrf2 is a transcription factor responsible for cellular defense against the cytotoxic effect of oxidative stress.  Under normal condition, Nrf 2 is kept in cytoplasm by Keap1 protein, which leads to Nrf2 degradation, and the half-life of Nrf2 is about 20 minutes. Oxidative stress results in dissociation of Nrf2 from Keap, and then Nrf2 translocates to the nucleus, where it binds to the specific DNA sequence - the antioxidant response element (ARE) in the promoters of its target genes, including Heme oxygenase (HO-1) and glutathione S-transferase (GST).  Because of its importance in regulating oxidative stress, Nrf2 is believed to be a good drug target candidate in the prevention and treatment of cancers.
