miRNA Plate Assay Kit MA-0101


MicroRNAs (miRNAs) are small noncoding RNAs that control gene expression at the posttranscriptional level through selectively binding to complementary messenger RNA sequences.  Approximately 30% of mammalian genes are regulated by these small RNA molecules, which affect a variety of biological functions, including development, cell differentiation, proliferation, apoptosis, and maintenance of stemness and imprinting. 
Northern blotting has been the most commonly used method for analyzing individual miRNAs, although recently a number of new approaches have been developed, such as the related real-time PCR and invader assays. Low sensitivity and poor throughput are the main issues of Northern blot analysis. To resolve this problem,  Signosis has developed the miRNA plate assay kits. The kits are highly sensitive, and the biochemical conversion of miRNA molecules into cDNAs is not needed. The procedure of the assay is as simple as incubation and wash.

To analyze a  miRNA with the plate assay, you need to purchase the common component miRNA plate assay kit (MA-0101) and  a specific oligo mix (MO-XXXX) corresponding to the miRNA.

miR-21 Plate Assay Kit MA-0104


miR-21 is overexpressed in a wide variety of cancers. It is believed to be an oncogene by downregulating the expression of many tumor suppressors, including programmed cell death 4 (PDCD4), tumor suppressor Pdcd4, the PTEN tumor suppressor gene, and the tumor suppressor gene tropomyosin 1 (TPM1). miR-21 is also found to be an important regulator of cancer-cell survival. Different from the survival pathway mediated by IL-6/Stat3 activation through the expression of antiapoptotic proteins, miR-21 is upregulated by Stat3, which mediates the suppression of apoptosis possibly through the inhibition of TPM1 and/or other proteins. Signosis’ miR-21 plate assay kit provides a simple and quick method to analyze this miRNA molecule.

miR-17-92 Cluster Plate Assay Kit MA-0102


A polycistronic miRNA cluster miR-17-92 plays a role in the control of cell proliferation and angiogenesis. This cluster consists of seven miRNAs: miR-17-5p, miR-17-3p, miR-18a, miR-19a, miR-19b, miR-20a, and miR-92. Like E2F1 gene, the cluster is transcriptionally activated by the proto-oncogene c-Myc. Because two of the expressed miRNAs, miR-17-5p and miR-20a, repress the translation of E2F1, the level of E2F1 protein is modulated by this feedback mechanism.  Therefore, the accumulation of excessive amounts of E2F1 can be prevented and the states of cells towards apoptosis or cell division can be controlled. In addition, c-MYC-mediated induction of the miR-17-92 cluster results in down-regulation of the anti-angiogenic thrombospondin-1 and related proteins, such as connective tissue growth factor 2; Therefore, it promotes angiogenesis.  Furthermore, a recent study indicates that the cluster is significantly up-regulated at the clonal expansion stage of adipocyte differentiation. The miR-17-92 cluster has been implicated as the oncogenes in numerous tumor types.  Effective monitoring of the expression of the clustered miRNAs can help us better understand how the alteration of the cluster contributes to development of cancers and shed light on the molecular mechanisms of miRNA function.

Stem Cell-Specific miRNA Plate Assay Kit


Some of the miRNAs appear to be stem cell specific, and microarray analysis of miRNA expression can profile a set of unique miRNAs in stem cells. Signosis developed a panel of miRNA plate assay kits to analyze these miRNA molecules. Each kit can be used to profile 8 stem cell related miRNAs.

Cancer miRNA Profiling Plate Assay I MA-0103


miRNAs have been implicated in the regulation of various biological processes such as cell death and cell proliferation. They can act as oncogenes and tumor suppressor genes, playing a crucial role in tumorogenesis. The change in the expression level of miRNAs is associated with dysfunction of their corresponding protein-coding gene targets, many of which are involved in the initiation and progression of cancer. For example, BCL2 oncogene is targeted by miR-15a and miR-16, while PTEN tumor suppressor is targeted by miR-21, and HOXD10, Pak, MYOD1 and ER, and Her2 are targeted by miR-10b, miR-7, miR-206, and miR-125a, respectively.
