GAS Luciferase Reporter HCT116 Stable Cell Line is derived from the human colon carcinoma cells (established from the primary colon carcinoma of an adult man). The cell line expresses the firefly luciferase reporter gene under the control of three copies of GAS (interferon gamma-activated site). This cell line is an ideal cellular model for monitoring the activation of IFNγ/JAK/STAT1 signaling pathway triggered by stimuli treatment, enforced gene expression, and gene knockdown.

 

IFN-γ (interferon-gamma) plays a pivotal role in regulating immune response and cancer. Upon binding IFN-γ to its receptor (IFNGR complex) it will induce JAK1 and JAK2 phosphorylation, which then allow binding of STAT1. Then this phosphorylated, homodimerized complex will translocate to the nucleus, where it binds to GAS elements within the promoter of IFNγ inducible genes.

To monitor and measure STAT1 activity upon IFN-γ induction we have generated “GAS Luciferase Reporter HCT116 Stable Cell Line”. The stable cell line has been made by transfection of a plasmid containing three copies of GAS (interferon gamma-activated site) located at upstream of a firefly luciferase gene.

 

Principle behind TF luciferase reporter. TF luciferase reporter stable cell line utilizes artificial promoter constructs to drive luciferase expression. The promoter region can consists of multiple repeats of a cis-element TF binding site, a DNA fragment from the promoter region of a known TF downstream gene, or a DNA fragment containing putative/known TF binding sites. There are several ways that a TF can be activated, such as through extracellular stimuli or through intracellular signaling pathways. Once activated, the TF translocates to the nucleus and often interacts with relevant co-factors to drive gene expression. Once luciferase is expressed, it can generate light in an enzymatic assay and the amount of light measured is positively correlated with the level of TF activation.
