Principle
Signosis’ Multi-Lipid Detection Kit provides a convenient and efficient multiplexed solution for the simultaneous quantification of three essential lipid species: triglycerides, total cholesterol, and free fatty acids. This assay utilizes specific enzymatic reactions for each lipid type to generate a measurable colorimetric product, via H2O2 detection, that quantifies the lipid levels in each sample. The resulting signals are easily measured using a standard microplate reader, enabling high-throughput lipid profiling in biological samples such as serum, plasma, or cell lysates.

Triglycerides – enzymatic hydrolysis and oxidation produces H2O2 which is detected with HRP.

Cholesterol – both free and esterified forms can be measured using cholesterol oxidase and cholesterol esterase, which generates H2O2 that can be detected with HRP.

Free fatty acids – enzymatic activation into fatty acyl-CoA and subsequent enzymatic oxidation produces H2O2 that can be quantified through HRP detection.
