TF ELISA Kits

Signosis’ highly sensitive Transcription Factor (TF) ELISA Kits provide a reliable and efficient method for detecting low levels of activated transcription factors in nuclear extracts. Compared to traditional techniques like EMSA or supershift assays, these kits offer a faster, easier workflow without compromising sensitivity—making them ideal for routine screening and high-throughput applications.

Principle

The Signosis TF ELISA Kit is designed to detect activated transcription factors in nuclear extract or whole cell lysate using a plate-based, antibody-mediated detection system. The 96-well (12×8) strip plate is pre-coated with a double-stranded DNA oligonucleotide containing the consensus binding sequence specific to the target transcription factor (TF).

When the sample is added, activated TFs bind to the immobilized DNA. A primary antibody specific to the TF subunit is then applied to detect the bound TF, followed by an HRP-conjugated secondary antibody. Upon addition of TMB substrate, a colorimetric signal is generated, which is directly proportional to the amount of activated TF in the sample. The signal is measured using a standard microplate reader, providing a sensitive and quantitative assessment of TF activation.

Benefits

Positive Control

An active recombinant protein with DNA binding activity is included in the kit as a positive control.

Singleplex and Duplex Formats

Can detect up to two TFs with one ELISA kit.

Highly Specific

Determines the activation of a specific TF gene using its DNA binding sequence and a specific antibody against the TF.

High-Throughput

Cell lysates from a 96-wel tissue culture plate can be prepared with Signosis' Whole Cell Lysis Buffer and analyzed with the TF ELISA kit.
