Principle
Signosis' TF Gaussia Luciferase Reporter Stable Cell Lines utilizes a specially engineered promotor construct to express Gaussia Luciferase under the activation of your desired TF of interest. The promoter region consists of multiple repeats of a cis-element TF binding site, a DNA fragment from the promoter region of a known TF downstream gene, or a DNA fragment containing putative/known TF binding sites. Once activated, the TF translocates to the nucleus and often interacts with relevant co-factors to drive Gaussia Luciferase gene expression.
The Gaussia luciferase reporter is derived from the marine copepod Gaussia princeps, which is a naturally secreted protein with a molecular weight of approximately 20 kDa and exhibits exceptionally high luminescence intensity. Its secreted nature allows the reporter enzyme to accumulate directly in the culture medium, eliminating the need for cell lysis during assay procedures.
Gaussia luciferase catalyzes a bioluminescent reaction using a coelenterazine-based substrate to generate light output.
