
Liver Enzyme Assays
Principle
Signosis’ Liver Enzyme Assay Kits provides a convenient and efficient multiplexed solution for measuring the activity of four key liver enzymes used to evaluate liver function in patients: alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), and gamma-glutamyl transferase (GGT).
These assays utilizes substrates to react with each liver enzyme and generate a colorimetric signal that can be measured with a plate reader, enabling high-throughput profiling in biological samples such as serum, plasma, or cell/tissue lysates.

ALT– ALT transfers an amino group from L-alanine to alpha-ketoglutarate, producing pyruvate. ALT activity is measured by quantifying the pyruvate using an enzymatic H₂O₂ generation reaction.

AST – AST transfers an amino group from L-aspartate to alpha-ketoglutarate, producing glutamate. AST activity is measured by quantifying the glutamate using an enzymatic H₂O₂ generation reaction.

ALP – ALP hydrolyzes p-nitrophenyl phosphate (pNPP) into pNP, releasing a yellow-colored product that can be measured at an absorbance of 405 nm.

GGT – GGT hydrolyzes γ-glutamyl-p-nitroanilide (GPNA) into pNA, releasing a yellow-colored product that can be measured at an absorbance of 405 nm.
