Principle
How the Competitive ELISA Assay Works
Unlike a standard sandwich ELISA, the signal in a competitive assay is inversely proportional to the hormone concentration.
1. Plate Coating: The 96-well plate is pre-coated with a target hormone-protein conjugate.
2. Competition: When your sample is added, the free hormones compete with the plate-bound hormones for binding sites on a biotin-labeled anti-hormone antibody.
3. Detection: After washing away unbound materials, Streptavidin-HRP is added to bind to the biotinylated antibodies immobilized on the plate.
4. Colorimetric Result: TMB substrate reacts with the HRP to produce a blue color, which turns yellow upon adding a Stop solution.
5. Quantification: Absorbance is measured at 450 nm. A stronger signal indicates a lower concentration of the target hormone in your sample.
The assay can be used to analyze serum, cell, tissue, and other liquid samples from human, mouse, and rat models.
