The NFκB Responsive Luciferase Reporter LNCaP Cell Line is derived from human prostate adenocarcinoma cells and stably expresses a firefly luciferase reporter gene under the control of an NFκB response element. This stable reporter line provides a sensitive and quantitative platform for monitoring NFκB signaling dynamics in a physiologically relevant human prostate cancer background.

 

The Advantage: Overcoming Transfection Bottlenecks

LNCaP is a notoriously difficult-to-transfect cell line. Generating stable, reliable reporter expression in this model typically requires weeks of tedious optimization, high failure rates, and wasted reagents. To solve this bottleneck, Signosis has leveraged a high-efficiency lentiviral transduction system to integrate the NFκB reporter stably and uniformly into the LNCaP genome. We have eliminated the challenging up-front optimization so you can immediately begin screening and generating data.

 

Biological Relevance

LNCaP is a widely used prostate cancer model valued for its sensitivity to androgens and intact androgen receptor signaling. In prostate oncogenesis, the NFκB pathway acts as a critical mediator of cell survival, inflammatory cross-talk, and progression toward hormone-refractory (castration-resistant) disease. This makes the line an ideal tool for investigating the intersection of inflammatory signaling and prostate cancer progression.

 

Mechanism of Action

NFκB is a master transcription factor regulating immune responses, cellular growth, and apoptosis. Upon activation by stimuli such as inflammatory cytokines, stress, or free radicals, NFκB translocates to the nucleus, binding to its response elements to drive downstream gene expression. Because aberrant NFκB activity is heavily implicated in tumor survival and therapeutic resistance, monitoring this pathway is essential for oncology drug discovery.
