The FKBP–FRB Positive Control Pair is a robust inducible protein–protein interaction (PPI) system designed to validate and optimize MoLuc PPI assays. The interaction between FKBP and FRB is specifically induced by rapamycin, making it a well-established positive control for split luciferase-based PPI detection.
In the MoLuc assay, FKBP and FRB are fused to the two complementary MoLuc luciferase fragments. Upon the addition of rapamycin, FKBP and FRB rapidly dimerize, bringing the luciferase fragments into close proximity to reconstitute an active enzyme that generates a bright luminescent signal.
This inducible system provides a reliable and reproducible method for confirming assay performance, optimizing experimental conditions, and validating reagent functionality.
The FKBP–FRB Positive Control Pair is available as plasmids for transient transfection making it an ideal control for assay development, quality control, and high-throughput screening applications.
