Signosis has established a human JAK2 WT (wild-type) + EPOR -expressing Ba/F3 cell line that serves as a valuable model for studying hematopoietic cell signaling, particularly the Janus kinase 2 (JAK2) pathway. JAK2 is a key tyrosine kinase involved in regulating fundamental cellular processes, including proliferation, survival, and differentiation of hematopoietic cells. This cell line functions as baseline control for investigating the physiological functions of JAK2 + EPOR in Ba/F3, comparing wild-type and oncogenic JAK2 variants, and evaluating the functional effects of mutations such as JAK2 V617F in related cell models.

 

The cell line was generated by lentiviral transduction of Ba/F3 cells with a lentiviral construct encoding human JAK2 WT. Stably transduced cells were selected using hygromycin, and positive clones were identified based on GFP expression. The selected clone was validated by PCR followed by sequencing to confirm the presence of the human JAK2 WT sequence and by Western blot analysis using an anti-JAK2 antibody to verify JAK2 protein expression.
