The Wnt1 Overexpression HEK293 Stable Cell Line is a Wnt1 producer cell line engineered to stably express human Wnt1, providing a reliable and reproducible source of biologically active Wnt ligand for co-culture-based Wnt signaling assays.
Unlike reporter cell lines, this cell line does not contain a Wnt-responsive reporter construct. Instead, it functions as a ligand-producing cell that continuously expresses membrane-associated and secreted Wnt1. When co-cultured with TCF/LEF luciferase reporter cells or other Wnt-responsive target cells, Wnt1 binds to the Frizzled (FZD) and LRP5/6 receptors on adjacent cells, activating the canonical Wnt/β-catenin signaling pathway. The stabilized β-catenin then translocates into the nucleus, where it interacts with TCF/LEF transcription factors to induce reporter gene expression.
Because Wnt proteins are highly hydrophobic and require cell-associated presentation for optimal activity, direct cell-to-cell contact between the Wnt1 producer cells and the reporter cells provides a more physiologically relevant method of pathway activation than treatment with recombinant Wnt proteins alone. This co-culture system offers robust, reproducible, and biologically relevant activation of canonical Wnt signaling while minimizing variability associated with recombinant ligand preparation and stability.
