Signosis has established the Jak2 V617F + EPOR expressing Ba/F3 cell line that faithfully recapitulates both ligand-dependent and ligand-independent JAK2 signaling, closely resembling polycythemia vera. This line enables detailed mechanistic studies of JAK2-EPOR interactions, robust STAT activation, and full cytokine-independent proliferation, providing an optimized platform for therapeutic screening and oncogenic transformation research.

 

This cell line uses Hygromycin, G418, and GFP as selection markers and can be cultured without IL3.

 

The cell line was generated by co-transfecting 2 vectors: one vector expressing JAK2 V617F under the MSCV promoter with GFP and Hygromycin resistance, and another vector with EPOR with G418 resistance, in parental BaF3 cells. Single clones have been selected with hygromycin and G418 treatment and GFP expression, with expression tested via Western blot. The clone with the highest expression were selected and expanded to produce these stable cell lines.
