Signosis has established a JAK2 T875N-expressing Ba/F3 cell line that serves as an important tool for researchers investigating the molecular mechanisms of myeloproliferative neoplasms (MPNs) associated with JAK2 mutations. Through stable lentiviral expression of constitutively active human JAK2 T875N, this cell line enables the study of mutant JAK2 signaling, downstream STAT5 activation, and transformation-associated phenotypes, providing a useful model for investigating disease mechanisms and evaluating targeted inhibitors.
The cell line was generated by lentiviral transduction of Ba/F3 cells with a lentiviral construct encoding human JAK2 T875N. Stably transduced cells were selected using hygromycin, and positive clones were identified based on GFP expression. The selected clone was subsequently validated by PCR followed by sequencing to confirm the presence of the JAK2 T875N sequence and by Western blot analysis using an anti-JAK2 antibody to verify JAK2 protein expression.
