Signosis EL-010 is a stable Ba/F3 cell line expressing human EGFR with the A767_dupASV insertion in the loop following the αC helix of the kinase domain. This exon 20 duplication is commonly observed in clinical cases of non-small cell lung cancer (NSCLC) and is associated with resistance to certain EGFR tyrosine kinase inhibitors (TKIs).
 
The cell line was generated by lentiviral transduction of Ba/F3 cells with a lentiviral construct encoding the EGFR A767_dupASV mutant, together with an EGFP reporter to facilitate identification and selection of successfully transduced cells by FACS. Positive clones were further validated by PCR followed by sequencing to confirm the EGFR mutation and by Western blot analysis to verify EGFR protein expression.
