LIN28A is a developmental regulator that blocks maturation of the let-7 family of microRNAs, thereby maintaining stemness and promoting growth programs.
 
Binding property
LIN28A recognizes a GGAG-containing element within the terminal loop of pre–let-7 and related RNAs. Binding recruits terminal uridylyl transferases and prevents Dicer processing.
 
Assay notes
Uses a biotin-labeled RNA derived from the let-7 terminal loop containing the GGAG element. Ideal for studying miRNA biogenesis and stem-cell–like programs.
