Biological function
TTP is a zinc-finger ARE binding protein that promotes rapid decay of inflammatory mRNAs, such as TNFα and other cytokines. It is a key negative regulator of inflammation and immune responses.
Binding property
TTP recognizes class II AU rich elements, with a strong preference for UAUUUAU- / AUUUA-containing motifs in U rich context. Binding targets mRNAs for deadenylation and decay.
Assay notes
Probe is a biotinylated ARE containing canonical UAUUUAU motifs. The assay is sensitive to phosphorylation state and can be used to monitor TTP activation or inhibition in response to stimuli.
