By placing the luciferase gene under the control of native gene promoter regions, these constructs capture the natural arrangement of transcription factor sites, cofactors, and regulatory elements, delivering readouts that closely mirror endogenous gene expression rather than simplified synthetic motifs.
Principle

Unlike reporters built from repeated consensus binding sites, these promoter-based constructs integrate multiple upstream inputs and chromatin effects, improving specificity, reducing artifacts from promiscuous motifs, and providing transcriptional responses that more faithfully predict endogenous mRNA changes for each gene.
