WNT/β-Catenin Pathway

The Wnt/β-Catenin signaling pathway is a key regulator of embryonic development, cell proliferation, stem cell pluripotency, and tumorigenesis. Upon activation by Wnt ligands, stabilized β-catenin translocates to the nucleus and partners with TCF/LEF transcription factors to drive the expression of critical downstream target genes.

Signosis offers a comprehensive suite of validated TCF/LEF Luciferase Reporter and Wnt1 Overexpression Stable Cell Lines engineered to monitor, stimulate, and dissect Wnt pathway dynamics in live cells:

  • TCF/LEF Reporter Cell Lines (CHO-K1, HEK293, HeLa): Feature a reporter vector with 6 repeats of TCF/LEF binding sites driving firefly luciferase expression, enabling highly sensitive, quantitative measurement of β-catenin-dependent transcriptional activation in response to extracellular stimuli, gene knockdowns, or small-molecule modulators.

  • Wnt1 Overexpression HEK293 Cell Line: Stably overexpresses full-length human Wnt1 ligand, serving as a reliable endogenous source of active Wnt1 for paracrine signaling studies, co-culture experiments, and direct pathway activation.

  • Autocrine Wnt1/TCF-Luciferase Cell Line: Combines constitutive Wnt1 ligand production with TCF/LEF reporter readouts, establishing a self-activating autocrine signaling model ideal for high-throughput screening of Wnt/β-catenin antagonists without requiring exogenous ligand addition.
