The plenti-EGFR (wt) vector contains the wt EGFR gene under the MSCV promoter which can be used to infect both dividing and nondividing cells. It contains the HIV-1 LTRs and the lentiviral packaging signal (?) for retroviral expression, with specific elements (WPRE, cPPT, RRE) included for improved transgene expression. Downstream of the EGFR viii gene is a DYKDDDDK-tag for Western blotting. Furthermore, copGFP can be used as a selection marker when used with stable cell lines.
Epidermal growth factor receptor (EGFR) is a transmembrane tyrosine kinase receptor that plays a critical role in regulating cell division and death. The EGFR gene is present on chromosome 7p11.2 and has 28 coding exons in which exons 5�7 and 13�16 code for the ligand binding domain and exons 18�24 code for the TK domain.  Downstream effects of EGFR activation include modulation of three major pathways of RAS-RAF-MAPK, PI3K/AKT and JAK/STAT pathways which can lead to cell survival, proliferation and migration of cancer cells.
