Assay Kits
Signosis’ Mitochondrial Function Assay Kits provide a way to evaluate mitochondrial function by analyzing key mitchondrial enzymes and metabolites in cells or tissue samples. The Succinate Dehydrogenase (SDH) Assay measures the activity of SDH, an enzyme that plays a crucial role in both the Krebs cycle and the electron transport chain within mitochondria. The Caspase-9 Assay measures the activity of the mitochondrial caspase involved in triggering cell death. In addition to these enzyme assays, the Mitochondrial Function Combo Assay Kit also includes two other kits which measure critical metabolites in the mitochondria: ATP and NAD+/NADH. The ATP Assay is used to assess mitochondrial energy production, while the NAD+/NADH Assay is used to examine the redox balance in the mitochondria.
Principle
The Mitochondrial Function Assay Kits are designed to measure four key enzymes and metabolites in the mitochondria: succinate dehydrogenase, caspase-9, ATP, and NAD+/NADH. Each marker is detected using a distinct and validated method:

Succinate Dehydrogenase (SDH) activity is measured using DCPIP, a redox probe that changes color in response to SDH activity.

Caspase-9 is detected with a fluorogenic substrate that is specifically cleaved by caspase-9.

ATP is consumed by glycerol kinase to convert glycerol into glycerol-3-phosphate, which is oxidized to generate H₂O₂, detected with a fluorogenic probe.

NADH reduces WST-8 to a yellow formazan dye, which is measured by absorbance at 450 nm.
