Firefly–Renilla Dual Luciferase Reporter Vectors

Firefly–Renilla Dual Luciferase Reporter Vectors offer a powerful all-in-one solution for transcription factor (TF) analysis by combining two luciferase genes in a single plasmid. In these constructs, firefly luciferase is driven by a TF-responsive promoter containing an optimal consensus DNA binding sequence, enabling highly responsive and accurate quantitation of TF activity. Renilla luciferase is expressed from a constitutive CMV promoter to provide an internal normalization control. This design enables fast, accurate, and normalized reporter readouts in transient assays by correcting for variation in transfection efficiency, cell viability, or sample handling.

For most high-throughput or relative-induction experiments, untreated wells with the pTA-TF vector are sufficient as a baseline, while a pTA-control vector is only needed for precise measurement of basal promoter activity.
